KE00259 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The human Endostatin ELISA kit is to be used to detect and quantify protein levels of endogenous human Endostatin. The assay recognizes human Endostatin. An antibody specific for human Endostatin has been pre-coated onto the microwells. The human Endostatin protein in samples is captured by the coated antibody after incubation. Following extensive washing, another antibody of biotinylated specific for human Endostatin is added to detect the captured human Endostatin protein. For signal development, Streptavidin-HRP is added, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information
Endostatin, a 20-kDa C-terminal fragment derived from collagen alpha-1(XVIII) chain (COL18A1), is an endogenously produced inhibitor of angiogenesis. Many different proteases are able to cleave the NC1 domain of collagen alpha-1(XVIII) chain to produce endostatin including cathepsins L, B, and K, elastase. Endostatin inhibits proliferation and migration of endothelial cells, angiogenesis, and tumor growth. It may interfere with the pro-angiogenic action of growth factors such as basic fibroblast growth factor (bFGF/FGF-2) and vascular endothelial growth factor (VEGF).
