KE00285 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The human TK1 ELISA kit is to be used to detect and quantify protein levels of endogenous human TK1. The assay recognizes human TK1. An antibody specific for human TK1 has been pre-coated onto the microwells. The human TK1 protein in samples is captured by the coated antibody after incubation. Following extensive washing, another antibody of biotinylated specific for human TK1 is added to detect the captured human TK1 protein. For signal development, Streptavidin-HRP is added, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information
Thymidine kinase 1 (TK1) is a cytosolic enzyme involved in pyrimidine metabolism that catalyzes the addition of a gamma-phosphate group to thymidine and in regenerating thymidine for DNA synthesis and DNA damage. Among the four deoxyribonucleoside-specific kinases in mammalian cells, TK1 is the only one with the most restricted substrates specificity. TK1 has been applied as an important biomarker for the diagnosis of various cancers, including breast cancer, esophageal cancer, and lung cancer. TK1 upregulation was indicated as an early event in a study of breast cancer and further studies demonstrated a positive correlation between TK1 and cancer stage.
