Product Information
KE00480 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The human ATM ELISA kit is to be used to detect and quantify protein levels of endogenous human ATM. The assay recognizes human ATM. An antibody specific for human ATM has been pre-coated onto the microwells. The human ATM protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for human ATM is added to detect the captured human ATM protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information
The protein kinase ataxia-telangiectasia mutated (ATM) is named for the autosomal recessive disorder ataxia-telangiectasia (A-T), caused by loss of the activity of this kinase and characterized by a progressive loss of cerebellar neuron function resulting in debilitating ataxia,as well as dilation of blood vessels (telangiectasia). ATM is a 370 kDa highly phosphorylated protein which is constitutively expressed in numerous tissues. ATM is a master regulator of the DNA damage response, and it coordinates checkpoint activation, DNA repair, and metabolic changes in eukaryotic cells in response to DNA double strand breaks and oxidative stress.
