Product Information
KE00261 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The FAS/CD95 ELISA kit is to be used to detect and quantify protein levels of endogenous FAS/CD95. The assay recognizes human FAS/CD95. An antibody specific for FAS/CD95 has been pre-coated onto the microwells. The FAS/CD95 protein in samples is captured by the coated antibody after incubation. Following extensive washing, another antibody of biotinylated specific for human FAS/CD95 is added to detect the captured human FAS/CD95 protein. For signal development, Streptavidin-HRP is added, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information
Fas (CD95/APO-1) is a transmembrane glycoprotein belonging to the tumor necrosis factor (TNF) receptor superfamily. It can mediate apoptosis by ligation with an agonistic anti-Fas antibody or Fas ligand. Stimulation of Fas results in the aggregation of its intracellular death domains, leading to the formation of the death-inducing signaling complex (DISC). FAS-mediated apoptosis may have a role in the induction of peripheral tolerance, in the antigen-stimulated suicide of mature T-cells, or both.
