KE00246 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The Human IL-11 ELISA kit is to be used to detect and quantify protein levels of endogenous human IL-11. The assay recognizes human IL-11. An antibody specific for human IL-11 has been pre-coated onto the microwells. The human IL-11 protein in samples is captured by the coated antibody after incubation. Following extensive washing, another antibody of biotinylated specific for human IL-11 is added to detect the captured human IL-11 protein. For signal development, Streptavidin-HRP is added, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information
IL-11 is a multifunctional cytokine produced by a variety of cells, which regulates hematopoiesis, bone function and cytoprotecting in the gut. As a member of the glycoprotein (GP) 130 cytokine family, IL-11 can activate the JAK-STAT3 pathway. Its pleiotropic effect is based on both anti-inflammatory and pro-inflammatory effects. The anti-inflammatory effect is downregulating the macrophage function by inhibiting the IL-1β, IL-12 and NO production. The pro-inflammatory effect is inducing the initial CD4+ T cells to differentiate into Th17 cells, and causing the inflammatory response in the multiple sclerosis. IL-11 is related to inflammation and cancer since its transmembrane co-receptor IL-11Rα has been shown to overexpress in lung cancer, colorectal cancer, gastric cancer, breast cancer, prostate cancer and osteosarcoma.
