KE00501 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The human PINK1 ELISA kit is to be used to detect and quantify protein levels of endogenous human PINK1. The assay recognizes human PINK1. An antibody specific for human PINK1 has been pre-coated onto the microwells. The human PINK1 protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for human PINK1 is added to detect the captured human PINK1 protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information
The mitochondrial serine/threonine-protein kinase PINK1, also known as BRPK and PARK6, protects cells from mitochondrial stress-induced dysfunction. PINK1 performs many important cell functions and has been highlighted for its role in early-onset Parkinson's disease (PD). PINK1/Parkin-mediated mitophagy halts the accumulation of toxic mitochondrial products that can cause neuronal loss in PD and prevents damaged mitochondria from fusing with the healthy network. Moreover, both PINK1 and Parkin have also been shown to function in a quality control pathway parallel to mitophagy by facilitating piecemeal degradation of mitochondria via the formation of mitochondria-derived vesicles (MDVs).
