KE00358 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The human TREM2 ELISA kit is to be used to detect and quantify protein levels of endogenous human TREM2. The assay recognizes human TREM2. An antibody specific for human TREM2 has been pre-coated onto the microwells. The human TREM2 protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for human TREM2 is added to detect the captured human TREM2 protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information

TREM2 (triggering receptor expressed on myeloid cells 2) is a cell surface receptor belongs to TREM family that is expressed on osteoclast, dendritic cells, macrophages, nature killers, neutrophils and microglia. TREM2 is localized predominantly in the Golgi complex, but also shuttles to and from the cell surface in endocytic and exocytic vesicles. It associates with DAP12 to initiate the intracellular signalling cascade via an immunoreceptor tyrosine-based activation motif (ITAM) domain and tyrosine-kinases.
