Rat IL-6 Recombinant antibody, PBS Only (Capture)

Sandwich ELISA standard curve of MP00264-2, Rat IL-6 Monoclonal Matched Antibody Pair - PBS only. 83179-2-PBS was coated to a plate as the capture antibody and incubated with serial dilutions of standard Eg0800. 83179-3-PBS was HRP conjugated as the detection antibody. Range: 62.5-4000 pg/mL.

Three rats were given an intraperitoneal injection of 100 μg/kg of LPS. All ratswere killed 4 hours after receiving LPS. Spleens were harvested and broken into individual cells by using homogenizer. Ratspleen conditioned media wascollected after culturing in 60 mL of RPMIsupplemented with 10% fetal bovine serum and stimulated with 1 μg/mL of LPS for 18 hours. An aliquot of the cell culture supernatant wasremoved, assayed for rat IL-6 and measured 6,449.6 pg/mL.

One rat spleen was cultured for 18 hours in RPMI 1640 supplemented with 10% fetal bovine serum, 50 μM β-mercaptoethanol, 2 mM L-glutamine, and 100 μg/mL of streptomycin sulfate. The cells were stimulated with 10 μg/mL of PHA and 10 ng/mL of PMA. An aliquot of the cell culture supernatant was removed, assayed for rat IL-6 and measured 179.7 pg/mL.

Sixteen rat serum samples were evaluated for rat IL-6 in this assay. Fourteen samples were measured belowthe minimum standard of 62.5 pg/mL. The measured results of the two samples were 117.15 pg/mL and 133.51 pg/mL, respectively.
