Background Information
What is the molecular weight of IL-4?
IL-4 has a calculated weight of 14.9 kDa.
What cell type produces the IL-4 protein?
Mast cells in peripheral tissues produce the cytokines IL-4 and IL-13, both of which regulate response to environmental allergens. IL-4 in particular is synthesized rapidly following T cell activation to then stimulate downstream anti-inflammatory processes.
What activity does IL-4 stimulate?
IL-4 is a highly polyfunctional cytokine, contributing to the regulation of many cellular and immunological processes. Increases in IL-4 levels stimulate IL-13 production as well as helper T cell and B cell proliferative responses to anti-immunoglobulin antibodies. IL-4 also aids in controlling T helper cell type 2 (Th2) differentiation, driving differentiation of naive T cells to the Th2 pathway.
How do IL-4 levels respond to brain damage?
Ischemic neurons may secrete IL-4 to help decrease proinflammatory activity of macrophages.(PMID: 26269636)
How do IL-4 levels respond to allergens?
Asthmatics express stimulated IL-4 bronchoalveolar lavage during allergen challenges.
Can polymorphisms in the Il4 gene lead to asthma?
Polymorphisms resulting in mutant IL-4 proteins, are associated with asthma risk, development, and severity.
What does the IL-4: IFN-g ratio signify?
IL-4 promotes Th2 differentiation and stability, promoting its own production while inhibiting Th1 cell differentiation. The Th1 pathway, however, produces interferon gamma (IFN-g). The IL-4:IFN-g ratio gives indication as to which pathway is being enhanced or depressed.
