FH Martina (Verified Customer) (07-03-2023) | IF protocol for fixed cryosection of mouse skeletal muscle: after a quick permeabilization step with Triton 0,3% in PBS, block with BSA 0,5% and GS 10% in PBS for at least 2 hours at room temperature. Then incubate with primary antibody 1:100 in blocking solution overnight at +4°C. The day after, incubate with secondary antibody 1:200 in blocking solution for 2 hours at room temperature. WB protocol for mouse skeletal muscle proteins: block the membrane with 5% milk in TBS-T for at least 2 hours at room temperature; then incubate with primary antibody 1:1000 in 5% milk in TBS-T at +4°C overnight. The day after, incubate with secondary antibody 1:2000 in 5% milk in TBS-T for 2 hours at room temperature. Developing the membrane, you will see double bands between 39 and 28 kDa.
