Product Information

Proteintech's PR30004 (Mayer's Hematoxylin Solution) can be used for tissue and cell staining. Mayer's Hematoxylin can stain the cell nucleus to produce a blue color. The formula of this product has been optimized to avoid using toxic chemicals, e.g., mercury or methanol. The staining is simple and time-saving. 

The product is provided in a ready-to-use format. No further dilution is required. Each milliliter of this product can stain ~10 slides.

Usage

IHC counter staining:  

After immunohistochemistry signal development, apply the product to the tissue slides to make sure the tissues are fully covered. After staining for 1-2 minutes, rinse the slides thoroughly with TBST bluing buffer for 1-2 minutes, followed by immersing in distilled water for 2 minutes, or rinse the slides in distilled water, followed by immersing in a saturated Na2HPO4 solution for 2 minutes. Once the slides turn blue, they can be washed, dehydrated, cleared, mounted, and observed.

H&E staining: 

1. For FFPE samples: Immerse slides in xylene for 10 minutes. Repeat in fresh xylene again for another 10 minutes. Then rehydrate the slides by sequentially incubating in 100%, 95%, 80%, and 60% ethanol solutions for 5 minutes each. Finally, wash the slides with distilled water for 2 minutes.

2. Apply the product onto the tissue slides and make sure the tissues are fully covered. After staining for 1-2 minutes, rinse the slides thoroughly with TBST bluing buffer for 1-2 minutes followed by immersing in distilled water for 2 minutes, or rinse the slides in distilled water followed by immersing in a saturated Na2HPO4 solution for 2 minutes. Apply Eosin staining then. 

3. For frozen samples: Wash the samples with distilled water. Then, perform staining using the same protocol as above. 

4. For cell samples: Fix the cells with 4% PFA for 10 minutes or longer. After fixation, wash the cells twice with distilled water for 2 minutes. Then, stain with the same protocol as above. If the cells are embedded with paraffin, please de-paraffin using the FFPE sample protocol above.


Cautions

1. Stain with 1-2 samples to make sure the staining achieves optimal results. 

2. Wear a lab coat and gloves during operation.
