Product Information
KE10033 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The Mouse DKK1 ELISA kit is to be used to detect and quantify protein levels of endogenous mouse DKK1. The assay recognizes mouse DKK1. An antibody specific for mouse DKK1 has been pre-coated onto the microwells. The mouse DKK1 protein in samples is captured by the coated antibody after incubation. Following extensive washing, another antibody of biotinylated specific for mouse DKK1 is added to detect the captured mouse DKK1 protein. For signal development, Streptavidin-HRP is added, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information
Dickkopf 1 (DKK1), a foundational member of the Dickkopf proteins, has been identified as a naturally secreted glycoprotein that typically antagonizes the Wnt/β-catenin signaling pathway by sequestering the Wnt receptor complex LRP5/6 (PMID: 31335221). DKKs play an important role in vertebrate development, where they locally inhibit Wnt regulated processes such as antero-posterior axial patterning, limb development, somitogenesis and eye formation. In the adult, Dkks are implicated in bone formation and bone disease, cancer and Alzheimer disease (PMID: 22738799).
