Background Information
The TPMT gene is localized to chromosome 6p22.3 and is approximately 34 kDa in length. It has an open reading frame (ORF) of 735 bp that encodes for a 245-amino acid peptide. The wild type TPMT allele, expressing the enzyme with normal activity. To date, there have been 30 variant TPMT alleles described (PMID: 22493988). Thiopurine compounds are commonly used in the treatment of childhood acute lymphoblastic leukemia, and as immunosuppressants following organ transplantation or for treatment of various autoimmune disorders. Thiopurine S-methyltransferase (TPMT) is required for detoxification, through S-methylation, of 6-thioguanine nucleotides (TGNs), a byproduct of thiopurine metabolism. Catalog#10682-1-AP is a rabbit polyclonal antibody raised against the full-length of human TPMT. TPMT has a predicted molecular weight of 28 kDa, however, as a result of its phosphorylation it separates on a SDS gel as a band of approximately 28-32 kDa.
